I only know of the Eurolines company. You can check their routes, fares, times here:

http://www.touring.de/
is the main German company. The Czech Republic is 'Tschenien' even on the English pages. There seem to be two buses per day from Heidelberg, one in the early morning and one overnight. The website will give you more info, fares etc and you can book online.
I can't find another company which runs this route.

Why check that site?
The information there is exactly the same as that given in previous posts on this thread.
Travel Tips for Prague
Chech BEER - "PIVO"
by canaglic
Czechs amber liquid, called ‘Pivo’ is world famous, and for many centuries has been the finest beer available. The distinctive flavour of Czech beer comes from hand picked Bohemian hops. The Czech Republic is the largest consumer of beer in the world, beating Germany. That means that every adult drink on average 1L of beer a day. The majority of beers produced in Prague is lager, the best known being Pilsner Urquell and Budvar (Budweiser). Popular Czech brand includes Gambrinus, Staropramen, Krusovice, Radegast and Velkopopovicky Kozel. PILSNER URQUELL - The original Pilsner beer, came in an overwhelming first place, echoing a recent poll in which the majority of Czech citizens called Pilsner "the beer with the highest quality."
GAMBRINUS - Czech popular beer. Gambrinus pale lager is distinguished by its rich gold color and ample foam. Its outstanding feature is the full-bodied flavor with fine bitterness.
STAROPRAMEN - is another Czech popular beer exported to 36 countries. Pivovary Staropramen a.s. is the second largest beer producer in the Czech Republic, with a 14% share of the domestic market.
There is so much to see but it...

There is so much to see but it is almost all within walking distance. The public transport is also good and inexpensive. Try to go to Vysehrad (not sure of the spelling), there is the remains of a fort and it has a cemetary with various famous Czechs and a beautiful park, it is also out of the main tourist area a little and has a unique atmosphere. This is the Slavin Monument in the cemetery, at the base are my friends Marie, Ali and Pat.
Kneeling begging
by jo104
I noticed this a lot along the streets, people crouching down with their faces covered by hoods begging for money then next minute they are gone. Apparently they do not show their face so that people can give annonomously.
I am not a fan of people begging so I passed by.
Packing List
by Duffelgirl
Just the usual, The weather in March was fairly warm and I have heard it does get very hot in the summer...so plenty of shorts and t-shirts if you're planning a trip then.. I never found myself needing anything I couldn't easily find in a supermarket. Lots of film to get snap happy with. I never had a problem finding new films or batteries.
Prague: Graffiti Pt.1
by Intrepidduck
Written On the Wall - Golden Street.
If your on "Golden Street" high up in the Castle district of Hradcany you are sure to go to the Frans Kafka Museum. It's a tiny medieval house nestled between a few others which actually form part of the very castle's walls.
I've never actually been inside it, as it's been either too early in the morning or during the day there can been discovered a long queue, and often school groups, at the door. The former home of Prague's best known son has been visited, it is sure, by all the sons and daughters of Prague. In an unintentional ready-made artist installation the wall of one of the nieghbouring shops (former house) has become a wonderful scrawling of people's names and messeges. I'm sure that the shop's owners paint it over every month, but it's always there and will certainly stay as long as the sons and daughters of Prague come to visit the former home of their city's best known son.
Questions and Answers
Q: Prague public transportation "Hello VT;
I'm so confused how the 1 day or 3 days pass works. It says that you have to validate the pass/ticket before..."
A: "You validate it once and then it is good for the 24/72 hours on all public transport whether metro, bus or tram."
