Probably the best thing to do to get aquainted with the city is to take a walking tour of the city. There are many available and leaflets can usually be found in hotels and hostels plus the tourist information. You'll get to learn more about the history of the city and notice things that you otherwise would miss out on. From here you can then explore the city and points of interest in more detail.

Charles Bridge. Maybe the more beautiful bridge all over the world, its 30 statues, make the bridge a barroquian sculpture museum.
Do not forget touch John Nepomucenus statue if you want come back to Prague.
Ladies take your hairdryer

Take some comfy trainers as you will tend to walk a lot around Prague and a lot of it is cobbled and your feet will be in bits after a couple of days.
The weather is almost the same as the UK, I was out in September and it was mid 70's so take t-shirts and shorts for the day time and then casual stuff for the evening. Take a loo roll if you can because sometimes hotels will only give you one. You can get everything else you need in Prague. You will take a lot of pictures so make sure you take your phone/cam charger and a two pin adapter.
Mushroom picking just out of town

If you have a car there are many places just outside Prague that you can go mushroom hunting. One place, accessed by following the same road you would take to Kutna Hora, until you pass by a town called Mukarov..then head south into the reserve.
Mushroom picking is a very popular Czech activity, with most natives able to recognise the edible varieties. Even in late October you can find a basket full in a couple of hours...the difficulty being able to see them against fallen autumn leaves!
One danger though are the wild boar. They are huge beasts and they can be aggressive when they have young. You will see signs that they have been there even if you don't see the animals themselves. You will also notice the boar hunter huts.
Staropramen Brewery tour

The best beer comes from the Czech Republic, so why not go see how they make it? We visited Staropramen's factory. The tour includes: English speaking guides if requested, a video screening, historical info and viewing of old pieces of equitment, fermentation - & lager cellar and beer tasting.After the tour, you will get a HUGE glass of beer, about ½ a litre! The tour was quite short and it'd be nice if we could had gone around and looked by ourselves. Still, I'm satisfied with the tour.
