Can someone help with the best way to get from Rome to Alfedena?

www.viamichelin.com will give you routes, info and estimated timings.
By train?

will give you times, details and fares in English. Takes 3+ hours, with a change, one-way fare from 11.70 euro.

"Best"? We need some more parameters, like your expenses account, timetable, time of day you need to be there, all sorts of things.
Note that there is rail service to Alfedena, although not frequent. Go to www.trenitalia.com, look for English at the top, and enter "Rome" for the starting point and "Alfedena" for the destination, then enter a date in the next week that is the same day of the week that you intend to make the journey.
You'll note that the desitnation is "Alfedena-Scontrone", the next town up the line. Well, wherever the station is, there is likely bus service in time with the train to get you to Alfedena, if the stop is away from Alfedena.

arr Alfedena-Scontrone - 16:54
just 11.70 euro
There might be direct buses, but I wouldn't hold my breath - Alfedena is pretty small...
