First question: where will you be based/flying into?
Second question: how will you be getting around?
leics' question is very appropriate. As a matter of facts, you will move much more easily by car than by public transport, especially in Umbria.
I have just been in Tuscany and visited San Gimignano (UNESCO), Volterra (Etruscan town), Poggibonsi (where we stayed at the hotel), Monteriggioni (still fortified town), Colle Val D'Elsa and Certaldo (Boccaccio's hometown).
You probably won't be able to see all of them, but I suggest you'd go to Volterra at least. San Gimignano is very touristy, so I didn't like it that much even if it is very beautiful.
In Umbria, you should also consider Spoleto, at least.
Gubbio, Spello and Trevi, in addition to Michele's suggestion for Spoleto.
will fly to rome. I havent booked accommodation yet, but i think we can stay in 2 or more places -i dont want to spend many hours in the car...
i am also quite into wine. But i know that i have limited time. AAre Montalcino and Montepulciano worth visiting and spending some hours at one or two wineries?
I suggest to consider Montepulciano as base because there are several places to stay and it is a superb location to visit what you are looking for:
siena one hour by car
Assisi one hour and 30
Perugia one hour
plus several other hill towns as Pienza, montalcino, Monticchiello, bagno vignoni
Where to stay: visit montepulciano.com
cioa and good luck
