Kelowna for a day
Gonna be heading for Vernon on a 3 day business trip...probably staying at the Best Western Kelowna Inn.
Should have one evening and one full day to myself for seeing the sights...what would you recommend? First time visitor and will have a car, early October.

The local Wineries will be in full swing.
The are some real gems try bcwine.com for much more info.
Most Wineries are a short drive from the city-if I was you I'd head North to sample some of the delights of cold climate wines.

Sit on a bench near Okanagan Lake with a Latte and watch for Ogopogo.
Some fabulous horseback riding and riding tours there to explore amazing scenery.

i'm confused, are you going to be staying in vernon or kelowna. two different cities?

I think I'll stay in Kelowna since I have an early morning flight out. Will drive to Vernon for business.

Here's what I like to do when I have a day in Kelowna...
Morning: Hop in the car and head downtown. Lots of neat little boutiques and shops to explore, or you can walk on the beach next to Lake OK.
Lunch: Doc Willoghbys (spelling!) is a great little bar and grill downtown. They have a patio on the street, and an awesome selection of beer on tap. The servers are really friendly, and Dave (the owner) is cool. Ask the bartender if you want to check out the night life... they should be able to let you know about any good live music and such.
After lunch: Pick a winery or two, and GO! Summerhill is one of my faves. They probably have the best guided winery tour. The wine is a little on the pricey side, but tasty! If you're there in October, wine season is full on, so you should be able to sample some great stuff. If you're peckish, have tapas on the patio and take in the view of the lake. There are a few other wineries nearby (less than 15 mins drive) so you can take in more than 1.
Hope this helps, Joe. Tourism BC has an office on the main strip in Kelowna. If you are staying in Kelowna (or Vernon, for that manner) the front desk can give you directions. The offices are great: tons of info, and the staff are usually locals well versed in what's going on.
Enjoy! The Okanagan is a great place to visit.
