What is the quickest way to make it from W. 45th @ 8th to the Carlyle Hotel (E. 76th) at 10:30PM?

at that hour, a taxi is by far the quickest and easiest way. by subway it takes a change somewhere on the east side at 53 or 59 St to the 6 train which stops at 77 and Lexington, leaving you with a four blocks walk. the taxi will move much faster than during the daylight hours.

Taxi. But it sounds like you're coming out of a show, in which case it'll be nearly impossible to get a taxi. You can take either the uptown M104 or M10 bus that run along 8th Ave. If you're on the M104, get off at 79th & Broadway and transfer to the M79 eastbound crosstown bus to 79th & Madison. If you take the M10, get off at 81st & Central Park West and transfer for the same M79 bus. (It still stops at 79th & Madison.)

You can also go by subway. Get the uptown E on 8th Ave. and 42nd and get off at Lexington/53rd, where you can transfer to the uptown #6 to 77th St. (The subway is safe, even very late. Your biggest worry is getting a seat!)
