Bus from Ambon (ferry included, leaving very early morning) to Saka, then by boat to Sawai. The buses are still running but the boat you have to charter now. The boat ride is quite nice but only reasonable if you are a big group.
Better take from Ambon a Bemo / Ankot (shared minibus) to Tulehu, 10000 Rupies and 40 minutes, from there a speed boat, 120000 Rupies and 2 hours, then a Bemo to the market for 5000 Rupies. Then a Kijang (shared taxi) from Masohi (Amahai) to Sawai which is either 100000 or 150000 Rupies and 3-4 hours, leaving from near the market and bus terminal on the main road (100 meter north of the small shopping center there).

Use only one to Sawai not to Wasai! You will have still 5 km to walk from the turnoff. They usually leave sometime after midday when full.
For this trip it is worth spending a bit more to have the car for yourself as it goes thou a marvelous scenery and you might want to stop sometimes.
