To get to Ueda from Nagano via the expressway you should take a left off the Chuo Expressway onto the Joetsu Expressway after the Matsushiro Reststop. Continue along the expressway and get of at the second Junction for Ueda. From Matsumoto it is a little more tricky as there is no expressway. I don't recommend driving (especially in winter!!) but you can take a local train to Shinonoi Station and transfer to a Komoro/Karuizawa bound train.
Ueda has no local train/subway system so the best way to get around is by bus or taxi.

The Nagano Shinkansen has a stop in Ueda (one stop from Nagano Station). It takes 30 mins from Nagano and 2 hours from Tokyo.
The Shinkansen is by far the most convenient way to travel around Japan. Unfortunately it isn't very cheap! Luckily there is a Japan JR Rail Pass available to foreign travellers. See the JR East website for details:
It must be bought abroad and stamped in Japan before you can use it. There are several options including one week (unlimited JR travel except for the super fast Nozomi service), two week, JR East and JR West passes. Check with your local travel agent for the best option or click on the link to see for yourself!
