Daily buses leave Karachi for Thatta. However in my opinion this is a road trip best enjoyed if you have your own car, or rent a van/car. It is a straight - properly carpeted road that takes you to Thatta from Karachi. It takes approx. 2 hours to reach; depending upon how many stops you make on the way for sightseeing.
Beware of overtaking buses/trucks as well as speed breakers when entering small hamlets/villages/cities.

For Car rental, the rates vary depending upon the vehicle, provider, and condition. You should expect around USD200 ~ USD 400 for whole day rental (I m not very sure about it since I drove by my own car).
For your own car, it may not be as expensive and the cost will be approximately USD40 for the fuel and etc. for the whole drive.

Bus fare in quite nominal of less than Rs.150 (approx. USD.3) one side, yet the journey may be exhausting and uncomfortable.
For bus, go to Malir or Qaid Abad stop and take any bus that goes either to Thatta or Hyderabad.

Thee are only two ways you can go to Thatta and adjacent areas. These are either by public transport using bus or hiring van, car for a day with or without driver (self-drive).
National Highway, N-5 is two lane road in fair, if not good, condition and you can travel at the speed between 60 ~ 90 kms/hrs depending upon the condition of road. For return, if taken on Super Highway (M-9), the road condition is very good.
