Thai Airways has a daily flight to and from Phuket. They have very nice aircraft considerign it is only a domestic flight. Their inflight snack is really yummy esp the Blueberry Muffin :-)

Budget Airline from Singapore that makes travelling affordable!
I paid $150 per pax including of all taxes... Now, where can I get that price for other airlines??
And mine you, that's not the cheapest ticket u can get to Phuket! You just got to wait for the right promotion at the right time....
Note: The leg space is abit too narrow but it's just a couple of hours journey so it shldn't bother you much...

If time is tight or money not a concern....fly in and our of phuket.....bangkok aiar has tons of daily flights to phuket from BKK and other places. They also have flights from Phuket to Samui (which is what we are doing here). Book enough in advance and a one way ticket is around $2000B. Flight time to and from is approx 1 hr....they do tend to run a little late at times.
Bus is great, train is better....plane is best.....mix it up and try them all.

There are now 5 airlines operating flights from Bangkok to Phuket. Thai Airways and bangkok Airways are full service airlines with flights from Bangkok to Phuket.
The budget airlines with flights from Bangkok to Phuket are Thai Air Asia, Nok Air and One-2-Go airlines. These Thailand domestic flights are good value whether flying with a full service airlines or a budget airline. The budget airlines flying to Phuket from bangkok offer cheap fares, the earlier you book in general the cheaper the air fare.

These 2 budget carriers operate flights between Singapore and Phuket: Jetstar Asia and Tiger Airways. Both airlines periodically offer great deals and promotions. I purchased my Singapore-Phuket tickets at SGD $9 per way (that's a total of SGD $36 for 2 persons, return flight tickets...about USD$20 or less!!!).
Being budget airlines, they don't serve meals on board. If you really want a drink and some snacks, you can purchase them from the flight attendants. Note that outside food is strictly not permitted on board!
Jetstar Asia have allocated seats, but not Tiger Airways.
You can book your tickets online. Very secure and convenient.

Air Asia has the best rates I have found in and around SE Asia and Thailand.
I usually fly last minute and 1-way (cause who knows where i'll want to go next) And have always gotten a great price with Air Asia.
Recently, I even booked the same day and got 1-way fare Phuket to BKK for $23 USD. the other low cost airlines are competitively priced as well, but, for me, Air Asia has larger routes (more cities serviced). I usually book online and they do offer better prices with advanced purchase. However, if and when you book the same day as travel or within 24 hours, you cannot do it online and must reserve a ticket on the phone and then pay for it at the ticket counter at the airport before check-in (2 separate counters at BKK and SIN)
The planes are well kept and free seating (like SouthWest in the states). Other great low-cost airlines: Nok Air, Bangkok Air, Tiger Air and JetStar Air ( Phuket or Bkk to Singapore), One Two Go by Orient Thai Air. .... all are great low cost carriers in and around Thailand.

1 thing to watch out for - all the Asia carriers (Thai Air included) have a 15 kg luggage limitation and charge 30-60 BHT per kg over. So your luggage could be more expensive then your person. If you are returning to an area and do not need all your baggage check with your hotel or storage at the airport for leaving it behind. Luggage Storage at BKK is 90 BHT per day (can add up fast) and is located in the International Terminal.

Air Asia flies to Phuket daily (sometimes twice daily) from Kuala Lumpur and from Bangkok. The fares are amazingly low, but are however seasonal and subjected to surcharges. If you have to catch a connecting flight. Do not rely on this airline as delays are not uncommon. But hey its cheap! adjust your expectations accordingly. Transfers between Patong Beach and the airport are approx THB500 and the journey is about 40mins. Arranging one from the front desk of your hotel may costs more.
