Mount Isa is where the two Queensland outback routes of Greyhound meet: The route from Brisbane to Mount Isa, and the route from Tennant Creek to Townsville (or of course vice versa). Thus, it is a good place to change busses, and have a stop there as well to see the town.
The ride from Brisbane to Mount Isa is 26 hours in total, so usually if you are travelling here you have a break in one or more of the towns in-between, for example Longreach or Winton, where I stayed, or maybe Charleville, Barcaldine or Cloncurry.
The ride from Tennant Creek to Mount Isa is eight hours, from Mount Isa to Townsville it is about twelve hours. Stops on the way might be Richmond, where I once worked, or Charters Towers.
On my second trip I went straight from Mount Isa to Townsville and then connected to Cairns (another six hours) - in Townsville you can connect north or south along the east coast.
If you go to Tennant Creek, you can connect to busses north to Darwin or south to Alice Springs and Adelaide.
Note that the Tennant Creek - Townsville service does not depart daily, but only four times a week.

Years ago Virgin Blue used to operate flights to Isa from Brisbane but due to the small population and tiny number of tourists, it's simply not profitable. So now only Qantas flies here and the airfare sky rocketed from around $100 to $200+ - $300 one way from Brisbane. Anyway, try your luck on their website. You might get lucky. Trick is don't buy your ticket too early. Usually the price will drop a week before the flight date, for a day or two, then bounce up again, so check everyday.

Travel to the surrounding areas is easily done by 2 wheel drive if you stick to the tar,if your wanting to go off road you should have a 4wd you dont want to get stuck out here believe me i got stuck in my 2wd and was sitting there for 6 hours before anyone came it was lucky i told them i was coming otherwise i couldve been stuck for days , be sensible choose the vehicle that best suits your activity .
