Kendall Airlines (subsidiary of Ansett Airlines) and Qantas both serve Hobart from Sydney and Melbourne. You can include Hobart with the various Air Passes that can be purchased from outside of Australia. Buying direct, at least a month in advance means good savings!
Walking is the best way to see the city but renting a car guarantees that you can see as much of Tasmania as possible. Bus routes are pretty good if you are back-packing.
