From the US, you can fly to Belize City and it takes about 6 hours, with connections in either Miami or Houston.
Bus is the way to go, unless you have lotsa $$ to take a charter plane. The buses are very organized and punctual. From Belize City to San Ignacio, it takes about 2 hours on the local bus (which is a school bus, an interesting ride; 2.50USD) or you can take the express bus for a shorter ride (luxurious air conditioned bus; 3.50USD).
Please note that Novello's Bus Terminal in Belize City has moved a few blocks from where it was originally, now it is right at the water canal (less than 5 minutes from its original location). It is a 15-20 minute walk from the Water Taxi Terminal. You don't really need to catch a taxi, unless you are lugging lots of luggage, or just feel lazy..
Here is a photo of the 'bus terminal' of San Ignacio. Be careful not to miss it, you won't see a building terminal, just an open lot, where the bus will stop, reverse and park for a bit.

Once you get off of a bus at the main station in Belize City, you will be swarmed with drivers trying to get you in their vehicles, some with various cars and others with vans. They all seemed relatively safe, as mine was. Just tell a driver where you want to go and if they know it. Most of the places in Belize City that travelers go seem to be known by the drivers. To go from to my hotel along the water a little bit out cost 7$BZ ($3.50US).
Also, from my hotel to the city center at the water taxi station, it cost the same amount of money.

You can take a city tour by taking the horse carriage. These are parked close to the Terminal Mall in Belize City.
