In general every point of interest is easily to be found in Litomysl, EXCEPT the Portmoneum, and therefore I think it is best to search for it like that :
You get out of the castle-park and always walk straight on in a street with high trees on the sides. You pass by the piarist church and the 2nd church and walk still in the same street until you get to a large building on the left with a tiny garden in front.
You go around this house, turn to the left and then you turn left again at the next possiblity.
You might expect a big building, infact it is just the small house on my picture and I also almost turned around before I took a closer look there.
You might also search for that wooden sculpture in order to get to Terzy Novákové 75

Taking the train is certainly the best , when you do not intend to visit other places in the area as well.
From Prague the train takes about 2,5 - 3 hours and you have to change trains once.
From Brno / Brünn the train takes about 3 hours and you have to change twice.
In gerneral trains in the Czech Republic are really cheap, but maybe not always as comfortable as in western Europe.

Be prepared for the fact that Litomysl is "off the beaten path" and driving there from Prague or Brno will take several hours, depending on deviations, trafic and all the other many things that make travelling by car in Czechia almost unpredictable.
When arriving from Prague it might be the best to combine a visit of Litomysl with a tour to Kolin, Kutna Hora & Sedlec bone church, Pardubice, Hradec Kralove, Chrudim.
Coming from Brno you may combine it with a visit in Zdar nad Sazavou & Zelena Hora, Lysice...
Litomysl is directely on the road #35 / E442 between Olomouc and Hradec Kralove. A small part of that road is in excellent condition, but most of it is rather small and bumpy...
