Assisi was always thought to be my first journey within Italy and, once here, it took me two years make the trip. It was worth the wait as I was able to see Assisi with a dear friend who always wanted to go as well.
It took an hour and a half - leaving Rome's Termini Station and changing trains in Foligno. We did not make reservations and there was no pressure to meet a schedule as trains leave every 45 minutes - miss one - take the next. Our train had the most beautiful interior and comfortable seats - in fact, the best train ride I have had in Italy. Returning direct on one train took almost 2 hours.
We purchased tickets through the automatic machines without waiting in lines. Remember to "validate" the ticket in the stamp machine prior to boarding the trains.
From Rome you will see Foligno, Spoletto, and Spello and much beautiful Umbria countryside.
Once arriving in Assisi, there are busses always available to take you up the hill to the entrance of Assisi.

If you're going to Assisi from South, for example from Rome, it is an excellent trip either by car or train. You will pass by Spello, Spoleto, and abundant beautiful countryside. There is ample parking in Assisi if you drive - if your choice is a rail trip, you won't be disappointed and there is great transportation from the train station in the Lower city to Upper Assisi.

On my second trip there, I took the train from Rome. The Eurostar train took about 2 hours on the way over, but on the way back I took a cheaper train that took a little longer (and had to switch trains in Foligno). It's easy to get train tickets either in Rome or Assisi, and not very expensive (around 9-15 Euros depending on which train you take).

The "Azienda Perugina della Mobilita" website [www.apmperugia.it] has the schedule for the "C" bus that goes between the Assisi train station and the old city

Note: you must buy a bus ticket BEFORE you get on the bus, because the driver does not sell them. They are sold in Tabbachi stores. There is one inside the Assisi train station.

We took train to Assisi from Perugia. Then an easy bus ride from the train station up to the town.

Linea C runs every 30 minutes.. once you step out from the train station.. look on left you will see the Fermata post..

When arrives in Assis, you can chose to either get off at Basilica of San Francesco d'Assisi or up to the hill till the final stop there (near Duomo).. We chose to visit Duomo first and let the bus do the climbing part..

At the front of Assisi railway station is the bus stop for Assisi town.If you pay the driver it is 1-50 euro , so buy a ticket at the newsagents on the station for 1 euro. It is 4 km to top, two main stops , first at the Basilica of St Francis and then the termius at top. My advise is go to top and work your way downhill till you come to the Basilica,then you can take the bus down.You have to have the correct change for driver At the station the bus goes the other way to the St Maria district to the church of St Maria del Angeli, well worth a visit.
