The yellow buses run frequently to and from Mellieha and unless you have a car they're the best way to get around.
Buses no. 44 and 45 go to Valletta, tickets cost 25c/person. These buses go via Mosta and it takes about 50-60 mins to reach Valletta, depending on the traffic. If you catch an older bus, the trip is not very convenient, but bearable.
Bus No. 645 goes to Sliema (via Paceville/St. Julian's), tickets 50c/person. More convenient route as it follows the coastline so you can enjoy the nice views on your way.

Billy's, on the main street, takes bookings for tours with Oasis (as well as providing taxis and car hire). They are reasonably-priced, and the three drivers we had were pleasant and knowledgeable.
The minibuses they use are fine in cooler weather but I imagine they'd be pretty uncomfortable on a hot day. Still, they get you to various places and give you some interesting titbits of information.
You can book lunch on some of the tours; it's up to you, but we didn't bother. Take a picnic though, because at least one of the restaurants they use (and it was very pleasant inside) is on an industrial estate. Nothing much there to see or do for an hour or so.

Wonderful buses on Malta. Route 45 (Valletta to Cirkewwa) was most useful for us, as it was running most frequently from where we were based, but you can also get the 43/44/50/441 and 145.
Times and routes vary (get a free bus map from the central bus station in Valletta to help you make sense of it all). Check the times if you are there out of season; some buses only run in summer.

Considering that the town is located on the way to Cirkewwa, the harbour to Gozo, it is pretty easy to get to Mellieha.
There are a few buses that stop to Mellieha:
No. 43: Mellieha - Valletta – Mellieha
No. 44: Mellieha Bay, Mellieha - Valletta - Mellieha, Mellieha Bay
No. 45: Cirkewwa, Mellieha Bay, Mellieha - Valletta - Mellieha, Mellieha Bay, Cirkewwa
No. 48: Cirkewwa, Mellieha Bay, Mellieha - Bugibba - Mellieha, Mellieha Bay, Cirkewwa
No. 145: Cirkewwa, Mellieha Bay, Mellieha - Valletta - Mellieha, Mellieha Bay, Cirkewwa (via Birkirkara Bypas and University)
No. 441: Mellieha Bay - Anchor Bay - Mellieha Bay
No. 645: Cirkewwa, Mellieha Bay, Mellieha - Sliema - Mellieha, Mellieha Bay, Cirkewwa
The prices of a ticket vary from 15 to 40 cents according to the route.

To travel to/from Mellieha, you can take the bus route Valetta – Mellieha – Cirkewwa (ferry to Gozo). From Mellieha Bay, it takes about 1 hour to Valetta. There’s also a bus route to Buggiba, and I think also one that goes from Mellieha Bay to Anchor Bay (Popeye Village). The buses go regularly, we never had to wait more than 15 minutes when we went direction Valetta.
There are the following bus routes:
Route 43: Valetta – Mellieha center
Route 44: Valetta – Mellieha center - Mellieha Bay
Route 45: Valetta - Mellieha center - Cirkewwa
Route 48: Bugibba – Cirkewwa
The best would be to get a map with all the bus routes. And ask the bus driver where he goes to so that you don’t find yourself in the wrong bus!
