Oldenzaal connects to the main Dutch railway network through Hengelo, to and from which there are trains every half hour with a journey time of ten minutes.
This section of railway is operated by the private train company Syntus and so the OV Chipcard is not valid for that part of any journey. Through tickets, either way, are perfectly good though.
From Hengelo there are local and intercity services to the rest of the country and international services into Germany and onwards to Szczezin in Poland.
The station is located about ten minutes walk from the town centre (which is well-signposted) and the local bus station (where many of the buses are also run by the Syntus company) adjoins that of the railway.
As with almost everywhere in the Netherlands there are loads of cycle routes around Oldenzaal and the Twente region in general. Maps of the main routes can be obtained from the VVV Tourist Office opposite the Basilica and cycles can be hired from the Siemerink bike shop on Steenstraat in the town centre.
The routes are well-signposted and are either colour-coded or/and numbered.

By bike! No problems with parking space and putting lots of money in the parking meter. Oldenzaal is only a small town, so the bike is a perfect way to get to the centre of town.

Oldenzaal easily can be reached by car via the A1-highway exits 32 & 33.
Exit 32 is at the West for the Hulsbeek recreational area.
Exit 33 is to the South for the Industrial area and the Twente Airport.
You can park on the street around the old center or in the basement parking garage of the Driehoek shopping center.
Oldenzaal can be reached by train.
From Hengelo you have to change trains to the local Syntus train.
