You can get the ferry from downtown Boston, at Rowe's Wharf, to P-town every day in the summer. The ride is pleasant {about 2.5 hrs, I think} and once you get there the beaches and Commercial St will keep you entertained for the day. The down side is that it is difficult to get an inexpensive room, especially w/o reservations. If you stay overnight, go to the Crown and Anchor and see the show.
Plimouth Plantation/Mayflower II
The plantation has a large 'community' filled with period houses and other buildings. The residents each have a story and will talk freely about their 'lives' and the community.
Go to the nearby Cape Cod region. You can indulge in whale-watching or just walk around. There's an island off Cape Cod that has a loooong bridge of stones connecting it to the mainland.
