Another option for getting around Paraguay and Asuncion is to hire a private driver. The advantage of hiring a driver is he has a lot of knowledge of the area, you can go where ever you want when you want. Basically you have a private tour guide. This option is probably not feasible for the solo traveler, but if you are going in a group I can highly recommend it. I have used the services of Carlos A. Fleitas Frisch many times when I am in Paraguay and have never been disappointed. He is very professional and knowledgeable of the area. As well as a very friendly person. The only obstacle is the lack of English.....so if you can't speak Spanish get you hand signals ready!
They drive like crazy people, you think when you first get there, then you get in to the rhythmic swing for the in and out choatic driving . The bus (no a/c)will get you were you want to go but its a little confussing so i recommend renting a car, you will be in heaven because of the a/c alone....lol, but be careful because the cows, bulls, chickens and dogs are on the loose, not so much in Asuncion but outside the city is where you need to be very cafeful.
Taking the city buses in an unfamiliar Latin American city is a real accomplishment, one that is actually possible in Asuncion, which isnt that big and not that easy to get lost in (they use a grid system). Buses cost about 2000 Guarani (like 25 cents) for anywhere, and if you ask anyone, they can easily tell you what goes where. Try it!!!!
Asuncion is very central in South America and one can travel to many destionations from the major bus terminal in the city. Prices vary quite a bit, the more expensive your ticket the more luxurious your bus. An example is the busride from Asuncion to Florianapolis Brazil. This is about a 20 hour bus ride that costs 150,000GS (approx 25US) on a modern greyhound type bus
